When I did this I used screw-top auto-clavable vials and agar to for slants.
For longer term storage I'd add sterile mineral oil to the slants (Also
autoclaved) into the tubes. By "autoclave" here I just mean pressure cooked
at 15psi.
Use parafilm to seal, then store each slant in it's own small baggie -- All
the slants were kept in a small tupperware container. refrigerated.
You should be able to get most of your supplies from Cymar or brewing places
that sell culturing stuff.
However -- over the long term -- I pretty much quit yeast ranching, because
ultimately it's a pain to periodically reculture, and strain drift is always
a worry. Also, it' *really* a pain to build up a pitching culture from a
slant prior to brew day.
I do still keep some of the "seasonal" strains around -- but overall, it's
easier just to pay White labs or Wyeast to deal with the hassles associated
with maintaining strains.
